چکیده

Introduction. Because of obvious concerns, it is not possible to thoroughly explore the adverse effects of anesthetics in children. However, the availability of neural stem cell derived models has provided an invaluable tool to examine the etiology of neurotoxicity associated with developmental exposure to general anesthetics. Stem cells and neurotoxicity. To evaluate the degree of anestheticinduced toxicity, cultured neural stem cells were examined, using specific biomarkers, to determine the effects of anesthetics on NSC proliferation/differentiation, the nature of anesthetic-induced toxicity, and possible mechanisms underlying anesthesia toxicity. Anesthetics and proliferation. Using recently developed methods, such as dynamic molecular imaging approaches, the relationships between anestheticinduced NSC damage and alterations in NSC proliferation have been addressed. Summary. This review focuses on how anesthetics direct or signal NSCs to undergo apoptosis or necrosis, how neuronal transmitter receptors affect neurotoxicity, and how NSC models may serve as the most expeditious platform for decreasing the uncertainty in extrapolating preclinical data to the human condition.

Alpha-Tocopherol increases the proliferation of induced pluripotent stem cell derived neural progenitor cells

In addition to its antioxidant effect, Vitamin E or α–tocopherol is suggested to enhance remyelination in the animal model of non-inflammatory demyelination. In this study, the possible proliferative effect of vitamin E on human- induced pluripotent stem cell-derived neural progenitors (hiPS-NPs) and the underlying mechanisms were investigated in vitro. NPs were induced from iPS cells via 3 ste...

The Proliferation Study of hiPS Cell-Derived Neuronal Progenitors on Poly-Caprolactone Scaffold

Introduction: The native inability of nervous system to regenerate, encourage researchers to consider neural tissue engineering as a potential treatment for spinal cord injuries. Considering the suitable characteristics of induced pluripotent stem cells (iPSCs) for tissue regeneration applications, in this study we investigated the adhesion, viability and proliferation of neural progenitors (de...

Advanced Pre-Clinical Research Approaches and Models to Studying Pediatric Anesthetic Neurotoxicity

Advances in pediatric and obstetric surgery have resulted in an increase in the duration and complexity of anesthetic procedures. A great deal of concern has recently arisen regarding the safety of anesthesia in infants and children. Because of obvious limitations, it is not possible to thoroughly explore the effects of anesthetic agents on neurons in vivo in human infants or children. However,...

Sevoflurane decreases self-renewal capacity and causes c-Jun N-terminal kinase–mediated damage of rat fetal neural stem cells

Increasing studies have demonstrated that sevoflurane can induce neurotoxicity in the developing brains. JNK normally promotes apoptosis. It was hypothesized that sevoflurane affected the proliferation and differentiation of FNSCs and induced cell apoptosis, which caused the learning and memory deficits via JNK pathway. Sevoflurane at a concentration of 1.2% did not induce damage on the FNSCS. ...

Monosialoganglioside 1 may alleviate neurotoxicity induced by propofol combined with remifentanil in neural stem cells

Monosialoganglioside 1 (GM1) is the main ganglioside subtype and has neuroprotective properties in the central nervous system. In this study, we aimed to determine whether GM1 alleviates neurotoxicity induced by moderate and high concentrations of propofol combined with remifentanil in the immature central nervous system. Hippocampal neural stem cells were isolated from newborn Sprague-Dawley r...
